Overview

Sprouty RTK signaling antagonist 2 (SPRY2) is an intracellular protein that modulates signal transduction pathways downstream of receptor tyrosine kinases, including those for fibroblast growth factor (FGF), epidermal growth factor (EGF), and hepatocyte growth factor (HGF)[1][5]. It functions primarily as a negative feedback regulator, attenuating cellular responses such as proliferation, migration, and differentiation by inhibiting the ERK/MAPK pathway[1][3][5]. SPRY2 contains a characteristic cysteine-rich domain enabling membrane localization and regulatory activity[1][5]. Its function is highly context-dependent, as SPRY2 can also potentiate RTK signaling (notably EGFR) by interfering with ubiquitin-mediated receptor degradation[1][3][7]. Aberrant SPRY2 expression has been implicated in multiple cancers (breast, liver, lung, prostate, lymphoma), where loss or silencing of SPRY2 may contribute to tumorigenesis[1][3][5]. In immune cells, SPRY2 modulates T cell activation and cytokine production and is linked to inflammatory responses such as asthma[7]. Despite its relevance as a modulator of oncogenic and immunological signaling, there are currently no approved direct pharmacologic modulators of SPRY2.
