SPRY domain-containing SOCS box protein 2 (SPSB2) is an adaptor protein characterized by a central SPRY (splA/ryanodine receptor) domain and a C-terminal SOCS (suppressor of cytokine signaling) box[1][6]. SPSB2 functions primarily as a component in a SCF-like ECS (Elongin BC-CUL2/5-SOCS-box protein) E3 ubiquitin ligase complex, mediating the ubiquitination and subsequent proteasomal degradation of specific protein substrates, most notably inducible nitric oxide synthase (iNOS/NOS2)[1][3][6]. By recruiting the E3 ligase complex to iNOS through its SPRY domain, SPSB2 helps limit nitric oxide (NO) accumulation and cellular toxicity in activated macrophages[2][3][6]. Genetic and biochemical studies indicate SPSB2 is an important negative regulator of innate immune signaling and may have relevance in modulating chronic or persistent infections by influencing host-pathogen interactions[2][3]. Alternative splicing of SPSB2 leads to multiple transcript variants, and the gene is located on chromosome 12p13[1].
