SQLE divergent transcript (SQLE-DT), also known as lnc030, is a long non-coding RNA highly expressed in breast cancer stem cells. It acts as a regulator of cell stemness and tumorigenesis by stabilizing the mRNA of squalene epoxidase (SQLE), thereby promoting cellular cholesterol synthesis. This activity upregulates the PI3K/Akt signaling pathway and supports cancer stem cell properties. SQLE-DT's mechanistic effect is mediated through interaction with the poly(rC) binding protein 2 (PCBP2), which serves as a bridge between lnc030 and SQLE mRNA. The lnc030-SQLE-cholesterol axis is considered a potential therapeutic target for breast cancer treatment.
