Squalene monooxygenase, encoded by the ERG1 gene in *Saccharomyces cerevisiae*, catalyzes the stereospecific oxidation of squalene to (S)-2,3-epoxysqualene, a key step in ergosterol biosynthesis. It is a flavoprotein monooxygenase containing FAD and utilizes NADPH and molecular oxygen for activity. Located primarily in the endoplasmic reticulum, this enzyme is a validated target for antifungal drugs like terbinafine. Inhibition disrupts cell membrane integrity in fungi.
