Overview

SR-related CTD-associated factor 1 (SCAF1) is a multifunctional nuclear protein that serves key roles in linking transcription by RNA polymerase II to mRNA splicing, by binding its C-terminal domain. SCAF1 is a member of the serine/arginine-rich (SR) superfamily of splicing factors and influences both pre-mRNA processing and transcriptional regulation. Beyond the nucleus, SCAF1 is involved in the assembly and organization of mitochondrial respiratory supercomplexes, thereby impacting cellular metabolism and energetic efficiency. The gene exhibits broad tissue expression and multiple splice variants, some coding and some non-coding, reflecting its regulatory complexity. SCAF1 has also been implicated as a tumor suppressor and potential cancer biomarker, particularly in pancreatic cancer, where its loss sensitizes cells to chemotherapeutics and modulates inflammatory pathways. While direct drug targeting or safety concerns remain unreported, SCAF1 is recognized as an important regulatory molecule with disease associations—suggesting it is a relevant molecular target for further study[1][2][3][5].
