The SRC family tyrosine kinases (SFKs) are a group of non-receptor protein tyrosine kinases that play critical roles in regulating cellular processes such as proliferation, differentiation, survival, migration, and immune responses. They are proto-oncogenes and have been implicated in various diseases including cancers and immune disorders. All typical SFKs share a conserved multidomain structure and are regulated by phosphorylation. SFKs participate in signaling pathways controlling diverse biological activities including gene transcription regulation, immune cell signaling, cell adhesion, cytoskeletal organization, cell cycle progression, apoptosis, migration, and transformation. Their dysregulation is often associated with oncogenic transformation due to loss of normal inhibitory control mechanisms.
