Src Homology 2 domain-containing protein tyrosine phosphatases (SHP) are a family of non-receptor protein tyrosine phosphatases that play critical roles in cellular signaling. The two main members are SHP-1 and SHP-2. These enzymes regulate a wide range of biological processes including cell growth, differentiation, survival, immune responses, and development. They modulate pathways such as RAS/MAPK/ERK for cell proliferation/differentiation; PI3K/AKT for survival; JAK/STAT for immune function; among others. Mutations or dysregulation of these phosphatases are implicated in various diseases like Noonan syndrome, Leukemia and Autoimmune disorders. Targeting aberrant activity with small molecule inhibitors is an area of active research due to their central role in oncogenic signaling networks.
