Overview

SRC kinase signaling inhibitor 1 (SRCIN1) is an adaptor protein that negatively regulates SRC family kinase signaling, mainly by activating C-terminal SRC kinase (CSK), which in turn inhibits SRC activity and downstream proliferative and migratory pathways[2]. SRCIN1, also known as p140Cap, SNIP, or KIAA1684, is involved in controlling postsynaptic actin cytoskeleton organization, cell adhesion, cell cycle progression, and cellular differentiation, and it plays roles both in cancer biology—regulating tumor growth, invasion, and metastasis—and in neural function, impacting synapse maintenance and neurotransmitter release[1][2][3]. High SRCIN1 expression can suppress tumorigenic features and enhance chemotherapy sensitivity in some cancers such as neuroblastoma and breast cancer, while in colorectal cancer, SRCIN1 appears upregulated and promotes malignancy, indicating tissue-specific functions[1][3].
