Overview

SREBF2 antisense RNA 1 (SREBF2-AS1) is a long non-coding RNA transcribed from the antisense strand of the SREBF2 gene locus. It is upregulated in various cancers, notably hepatocellular carcinoma, where its m^6A-modified form enhances SREBF2 gene expression by recruiting specific RNA-binding proteins (FXR1) and demethylase enzyme TET1 to the SREBF2 promoter, resulting in DNA demethylation and increased transcription. This promotes cancer cell proliferation, migration, and resistance to apoptosis, and confers resistance to the tyrosine kinase inhibitor sorafenib. Elevated SREBF2-AS1 levels (and its m^6A modification status) are associated with poor prognosis in liver cancer and may serve as a prognostic biomarker and candidate therapeutic target[2].

Mechanism of action

Epigenetic regulation: m^6A-modified SREBF2-AS1 recruits RNA-binding proteins (e.g., FXR1) and TET1 to the SREBF2 promoter, inducing DNA demethylation and upregulation of SREBF2 transcription[2]. Promotes proliferation, migration, and resistance to apoptosis partly by enhancing expression of SREBF2[2].
