SRY-box transcription factor 12 (SOX12) is a protein-coding gene belonging to the SOX (SRY-related HMG-box) family of transcription factors, specifically to the SOXC subgroup which also includes SOX4 and SOX11[1][2][4]. Members of the SOX family are characterized by the presence of a DNA-binding high mobility group (HMG) domain, which enables them to regulate gene expression by binding DNA and influencing chromatin structure. SOX12 functions primarily as a transcriptional activator, sometimes in cooperation with other transcription factors such as POU3F2/BRN2 or POU3F1/OCT6[1]. It participates in tissue differentiation, maintenance of cell types, organ development, and immune function through T cell differentiation[1][2]. Disruption or mutations in SOX12 are associated with certain developmental disorders, but, unlike some other SOX proteins, SOX12 knockout mice do not display strong phenotypes, likely due to redundancy with SOX4 and SOX11[2]. There is no current evidence supporting direct druggability or active therapeutic targeting of SOX12, although other SOX proteins are being investigated for roles in cancer[3][4].
