Overview

SRY-box transcription factor 13 (SOX13) is a member of the SoxD subfamily of the SRY-related high mobility group (HMG) box transcription factors. It functions primarily as a nuclear transcriptional regulator, binding to DNA at consensus sequences and repressing gene expression of target promoters, including those for myelin proteins and cytokines[1][2][4][5][6]. SOX13 modulates crucial developmental processes, including neuronal, mesenchymal, and chondrocyte differentiation, and is vital for T-cell subset differentiation, particularly gamma-delta T-cells and IL-17A-producing cells[1][2][3][4][5]. It is also involved in vascular biology, where flow-dependent SOX13 expression in endothelium suppresses pro-inflammatory cytokine and chemokine production; loss of SOX13 leads to enhanced endothelial inflammation, implicating it in atherogenesis[2]. SOX13 is recognized as islet cell antigen 12 (ICA12), and autoantibodies against it are associated with type 1 diabetes[1][2]. Its dysregulation is implicated in various diseases including diabetes, cancers, and vascular inflammation[2][3].
