SRY-box transcription factor 18 (SOX18) is a member of the SOXF transcription factor family responsible for controlling gene expression by binding to DNA through its highly conserved HMG box domain. SOX18 is crucial for embryonic development of blood and lymphatic vessels and continues to modulate endothelial cell functions in adult tissues. Its activity includes homodimerization and interaction with other transcription factors such as MEF2C and RBPJ. SOX18 is highly conserved among vertebrates and can regulate a broad array of genes through direct DNA binding and dimer formation, distinguishing its transcriptional signature from related factors like SOX7 and SOX17. Aberrant SOX18 expression or activity is implicated in angiogenesis-associated diseases, including various cancers and cardiovascular conditions. Pharmacologic targeting of SOX18—for example, by blocking its dimerization—can disrupt endothelial gene programs, suggesting a promising strategy for both anti-angiogenic and anti-tumor therapies. However, its widespread roles in development and vascular homeostasis present notable safety challenges.
