SRY-box transcription factor 30 (SOX30) is a member of the SOX (SRY-related HMG-box) family of transcription factors characterized by a DNA-binding HMG box domain[1][7]. SOX30 acts as both a transcriptional activator and repressor, binding to specific DNA motifs to regulate the expression of target genes. In mammals, SOX30 is essential for male germ cell development, being highly and specifically expressed in meiotic and post-meiotic testicular germ cells, especially during spermatogenesis[1][4][6]. Disruption or deletion of SOX30 in mice leads to impaired spermatogenesis, resulting in male infertility due to arrest during the transition from late meiotic to postmeiotic germ cell stages[1][4][6]. SOX30 additionally plays roles in the regulation of genes involved in cell fate determination and development, such as activating p53 transcription to promote tumor cell apoptosis in certain cancer contexts[7]. SOX30 can inhibit Wnt signaling by directly binding to and suppressing transcription of CTNNB1 (β-catenin), and by binding the CTNNB1 protein to block its interaction with TCF7L2/TCF4[7]. Its function is predominantly specific to the testis in adults, with limited expression in other tissues[1][5]. There are no approved drugs or therapeutic interventions targeting SOX30 directly, and it is not currently considered a conventional therapeutic target such as a receptor or enzyme.
