SRY-box transcription factor 4 (SOX4) is a member of the SRY (sex-determining region Y)-related high-mobility group (HMG)-box transcription factor family. SOX4 plays essential roles in embryonic development, stem cell maintenance, and cell fate determination[1][2][5]. It has a highly conserved HMG DNA-binding domain and acts as a key regulator of progenitor cell survival, inhibition of terminal differentiation, and modulation of multiple developmental signaling pathways, including Wnt/β-catenin signaling[4][1]. SOX4 is crucial for proper cardiac and B lymphocyte development—complete loss in mice results in embryonic lethality due to cardiac defects and blocked B-cell development at the pro-B stage[1][6][2]. In adults, it remains important for neural and immune system function and is implicated in various cancers, where its overexpression promotes tumorigenesis and cell stemness[1][2][4].
