Overview

SRY-box transcription factor 9 (SOX9) mRNA is the messenger RNA transcript encoding the SOX9 protein, a master regulator transcription factor containing a high-mobility group (HMG) DNA-binding domain (UniProt P48436). It plays a fundamental role in embryonic development, specifically directing chondrogenesis and male sex determination by regulating genes such as COL2A1 and AMH (NCBI Gene ID: 6662). In adult tissues, SOX9 mRNA expression is essential for maintaining the pluripotency and regenerative capacity of stem cell populations in the intestinal epithelium, skin, and liver (PMID: 28811286). Pathologically, the overexpression of SOX9 mRNA is a hallmark of several aggressive malignancies, including colorectal, lung, and breast cancers, where it drives tumor initiation, metastasis, and resistance to therapy by promoting the epithelial-to-mesenchymal transition (PMID: 30610214). Therapeutic targeting of SOX9 mRNA using antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs) represents a strategy to downregulate SOX9 protein levels and inhibit oncogenic progression (PMID: 24166171). However, because SOX9 is vital for normal tissue homeostasis and skeletal development, therapeutic interventions must carefully manage potential systemic toxicity and off-target effects (PMID: 25633101).
