SS18-like protein 1 (SS18L1, also known as CREST) is a calcium-responsive transactivator and an essential subunit of the neuron-specific BAF (nBAF) chromatin remodeling complex, homologous to the yeast SWI/SNF complex[1][3][2]. It acts as both a transcriptional coactivator and transcription factor. SS18L1 regulates the expression of neuronal genes by modulating chromatin accessibility in response to calcium signaling, in part by recruiting CREB-binding protein and other histone acetyltransferases to gene promoters[1][3]. Functional domains include an N-terminal auto-regulatory region, a central methionine-rich protein-interaction domain, and a C-terminal glutamine-rich transactivation domain[1]. It is predominantly expressed in neuronal tissue and Schwann cells and is implicated in the regulation of neuronal differentiation, dendritic growth, Schwann cell proliferation, migration, and differentiation in response to injury[1][3]. Mutations in SS18L1 are associated with amyotrophic lateral sclerosis (ALS) and related neurodegenerative diseases[1][3]. The protein interacts with multiple components of chromatin remodeling complexes and is essential for proper nervous system development and adult neurogenesis[1][3][5][6]. No approved drugs directly target SS18L1, and no established clinical biomarkers or targeted therapeutics are currently known for this specific factor[3].
