Overview

CLEVER-1 (Common Lymphatic Endothelial and Vascular Endothelial Receptor 1), also known as Stabilin-1, is a large multidomain transmembrane glycoprotein receptor expressed by subsets of vascular and lymphatic endothelial cells, as well as immunosuppressive macrophages and monocytes. It functions as a scavenger receptor, mediating endocytosis of various ligands, and plays a crucial role in leukocyte trafficking, particularly the migration of regulatory T cells. CLEVER-1 is involved in immunomodulation, with high expression marking anti-inflammatory macrophages. It is being explored as a therapeutic target in cancer immunotherapy, with monoclonal antibodies like bexmarilimab aiming to block its function and enhance anti-tumor immunity. High CLEVER-1 levels have been associated with poor prognosis in certain cancers.
