STAG3 cohesin complex component (STAG3) is a meiosis-specific structural subunit of the cohesin protein complex, predominantly expressed in primary spermatocytes and oocytes during gametogenesis[1][2]. Cohesin complexes containing STAG3 play essential roles in chromosome axis organization, homologous chromosome pairing, synapsis, recombination, and accurate segregation during meiosis[1][2]. As the main stromal antigen component in meiotic cells, STAG3 interacts with α-kleisin subunits (such as REC8 and RAD21L) and is required for the stabilization and functionality of meiotic cohesin on chromosome axes, contributing to both centromeric and axial cohesion[1][2]. Mutations or loss-of-function in STAG3 result in impaired synapsis, defective DNA repair, premature loss of centromeric cohesion, failure in homologous recombination, and early meiotic arrest, culminating in infertility due to defective gametogenesis[1][2]. STAG3 is not currently a therapeutic drug target, nor are any drugs known to target it for clinical purposes.
