Overview

STAM binding protein like 1 (STAMBPL1) is a zinc-dependent metalloprotease in the JAMM family, acting as a deubiquitinase with high specificity for Lys63-linked polyubiquitin chains. It regulates protein stability, degradation, and cell signaling—including the WNT/PI3K/NF-κB and TORC1 pathways—by removing ubiquitin from target proteins such as TRAF2 (tumor necrosis factor receptor-associated factor 2) and the EMT transcription factor SNAI1. STAMBPL1 is upregulated in several cancers, promoting cell proliferation, survival, and metastasis, and has been implicated as a novel biomarker and therapeutic target in oncology. Its depletion or inhibition can suppress EMT, enhance apoptosis, and reduce tumor growth, suggesting further therapeutic potential.
