Thermonuclease is a heat-stable, calcium-dependent endonuclease primarily produced by Staphylococcus aureus. It catalyzes the hydrolysis of both DNA and RNA at the 5' position of the phosphodiester bond, yielding 3'-mononucleotides. As a secreted enzyme, thermonuclease functions extracellularly and contributes to virulence by degrading neutrophil extracellular traps (NETs) and regulating biofilm formation. Two main forms exist: secreted Nuc1 and cell-surface attached Nuc2, each playing distinct roles in S. aureus pathogenicity. Its enzymatic activity is utilized for rapid identification of S. aureus in clinical diagnostics.
