Staphylococcus aureus capsular polysaccharide type 8 (CP8) is a crucial component of the bacterial capsule, contributing significantly to virulence and immune evasion. It is one of the two most prevalent capsule types found in clinical isolates of S. aureus. As an antiphagocytic factor, CP8 hinders recognition by phagocytic cells, promoting bacterial persistence. The trisaccharide repeating unit structure, consisting of N-acetyl-D-mannosaminuronic acid, N-acetyl-L-fucosamine, and N-acetyl-D-fucosamine, and its biosynthesis via the cap8 operon are critical for its biological activity. Due to its role in pathogenesis and the ability of anti-CP8 antibodies to promote bacterial killing, CP8 is a promising vaccine target.
