Overview

StAR-related lipid transfer domain containing 8 (STARD8) is a member of the Rho GTPase activating proteins family, with a characteristic lipid-binding START domain. It regulates GTPase activity (especially RHOA and CDC42), localizes to focal adhesions, regulates cell morphology, orchestrates membrane trafficking, and maintains organelle integrity. STARD8’s tumor suppressor function is indicated by inactivation mutations in multiple cancers. It plays developmental roles that are conserved across species, serving as a critical regulator of cellular architecture and signaling pathways implicated in cancer pathogenesis.
