Overview

STE20-related kinase adapter protein beta (STRADB) is a pseudokinase encoded by the STRADB gene, classified as part of the serine/threonine protein kinase STE20 subfamily; however, STRADB itself is catalytically inactive due to an altered active site residue[1][2][4][5][7]. STRADB functions as a regulatory adaptor/scaffold protein, most notably as a component of a complex that activates the master regulator kinase LKB1 (serine/threonine kinase 11), which is crucial for controlling cell polarity, energy metabolism, and enforcing G1 cell cycle arrest[1][2][4]. Through interaction with the X-linked inhibitor of apoptosis protein (XIAP), STRADB can enhance anti-apoptotic signaling via the JNK1 pathway[1][2]. While STRADB is encoded at a chromosomal region associated with juvenile amyotrophic lateral sclerosis (ALS2), its direct disease involvement is not definitively established[1][6]. There are no known drugs or pharmacological agents that directly target STRADB, and it is not currently considered a primary therapeutic target[2][7].
