STEAP3 antisense RNA 1 (STEAP3-AS1) is a long noncoding RNA located antisense to the STEAP3 gene. It is upregulated in colon and colorectal cancer tissues and cell lines and is associated with poor prognosis in these cancers[1][2][3]. STEAP3-AS1 exerts its bioactivity by modulating the expression and stability of the neighboring STEAP3 gene, which encodes an iron transporter. Mechanistically, STEAP3-AS1 can regulate cell proliferation, cell cycle, and cell migration by affecting downstream molecular pathways, including CDKN1C and Wnt/β-catenin signaling[1][2][3]. Under hypoxic conditions, HIF-1α increases the transcription of STEAP3-AS1, which then competitively binds the RNA-binding protein YTHDF2, stabilizing STEAP3 mRNA and promoting iron metabolism and Wnt/β-catenin signaling activation, thus contributing to cancer progression[2][3]. Knockdown of STEAP3-AS1 suppresses tumor growth, highlighting its therapeutic relevance as a molecular marker and a potential, although not yet direct, target in colorectal cancer research[1][2].
