Overview

Sterile Alpha and TIR Motif Containing 1 (SARM1) is an evolutionarily conserved enzyme that functions as an NAD+ nucleosidase. It plays a critical role in programmed axon death (Wallerian degeneration) by depleting axonal NAD+ upon activation. SARM1 is a therapeutic target for neurodegenerative diseases, and its inhibition has shown promise in preventing axon loss after injury or in disease. While it was initially studied for its role in innate immunity, its neuronal function is now considered more prominent. SARM1 is expressed primarily in neurons and also found in macrophages and T cells.
