Overview

Sterile alpha motif domain containing 4B (SAMD4B) is a widely expressed human protein composed of 694 amino acids, featuring a sterile alpha motif (SAM) domain that is crucial for RNA binding and transcriptional repression activity. Overexpression of SAMD4B suppresses the activity of major regulatory transcription factors including AP-1, p53, and p21, mainly via its SAM domain. SAMD4B regulates nuclear-transcribed mRNA poly(A) tail shortening and is active in the cytosol. In pathological contexts, particularly colorectal cancer, SAMD4B is a direct target of microRNA-451 (miR-451); decreased miR-451 results in elevated SAMD4B expression and is associated with increased tumor cell proliferation, migration, resistance to apoptosis, and reduced chemotherapy sensitivity, highlighting SAMD4B as a potential therapeutic target and biomarker for disease progression.
