Overview

Sterile alpha motif domain-containing protein 11 (SAMD11) is an evolutionarily conserved protein that functions as a transcriptional co-repressor through its role in Polycomb repressive complex 1 (PRC1)-like complexes, especially in retinal photoreceptor cells. It is predominantly expressed in developing and mature rod photoreceptors, where it interacts with other SAM-domain proteins to help maintain photoreceptor cell identity by repressing nonrod gene expression. In humans, SAMD11 also shows a broader tissue distribution and is involved in negative regulation of transcription mediated by RNA polymerase II. Mutations in SAMD11 are associated with retinal degenerative diseases such as retinitis pigmentosa, and the gene has also been implicated in neurodevelopmental disorders and cancer progression (through its circular RNA form, circSAMD11). No approved drugs are known to target SAMD11, nor is it a current focus for therapeutic intervention, but it may serve as a biomarker or research target for disease mechanisms involving epigenetic regulation and cell identity maintenance.
