Overview

Sterile alpha motif domain-containing protein 14 (SAMD14) is a member of the SAM domain protein family, whose domains are evolutionary conserved and known for roles in signal transduction, cell surface receptor activation, and transcriptional repression[2][1][3]. SAMD14 is distinguished by interactions with the actin capping protein complex (Capzβ, Capzα1, Capzα2), promoting stress erythropoiesis through regulation of actin dynamics and progenitor activity[1][3]. SAMD14 exerts functional effects on hematopoietic stem and progenitor cells, including both myeloid and erythroid lineages, with important implications for erythrocyte regeneration and cell survival under stress conditions[2][1][3]. Epigenetic silencing or downregulation of SAMD14 is associated with progression or poor prognosis in multiple cancers (adenocarcinoma, gastric cancer, prostate cancer), suggesting a putative tumor suppressor role[2][1][3]. In mast cells, SAMD14 modulates secretion profiles, influences ECM remodeling, cell adhesion, and immune regulation within the tumor microenvironment[2]. There are no reported drugs directly targeting SAMD14, nor specific safety concerns in the context of therapy noted in the available literature.
