Overview

Sterile alpha motif domain-containing protein 5 (SAMD5) is a protein characterized by the presence of a SAM domain, a 65–70 amino acid modular protein-protein interaction motif implicated in versatile cellular signaling and regulatory complexes. It is predicted to regulate intracellular signal transduction, with functional evidence supporting roles in inhibiting cell proliferation, cell invasion, and promoting apoptosis in various cancer models by downregulating oncogenic pathways such as c-Myc. SAMD5 acts as a tumor suppressor in breast cancer, especially in triple-negative breast cancer, counteracting the effects of oncogenes such as PLK1 and c-Myc, and its expression is notably reduced in cancer tissues compared to normal tissues. The protein localizes primarily in the cytoplasm but can also be present in the nucleus, particularly in cholangiocarcinoma cells, and may have broader but as-yet-unelucidated functions in hematopoiesis and epithelial tissues. There are no approved drugs targeting SAMD5, and it is not currently considered a direct therapeutic target in drug development pipelines.
