Overview

Steroid 17-alpha-monooxygenase, encoded by the CYP17A1 gene, is a crucial enzyme in steroid hormone biosynthesis. It catalyzes both 17α-hydroxylation and 17,20-lyase reactions, converting pregnenolone and progesterone into precursors for glucocorticoids and sex hormones. CYP17A1 is primarily expressed in the adrenal cortex and gonadal tissues. Its activity is regulated by cofactors such as cytochrome P450 oxidoreductase and cytochrome b5, as well as phosphorylation. Mutations in CYP17A1 can lead to various disorders, including congenital adrenal hyperplasia and disorders of sexual development. Targeted inhibition of CYP17A1 is used therapeutically in prostate cancer.
