Overview

Steroid receptor associated and regulated protein (SRARP) is a human protein encoded by the SRARP gene located on chromosome 1p36.13, consisting of 169 amino acids. SRARP is predominantly expressed in steroid hormone-responsive tissues and is co-expressed with the estrogen and androgen receptors in breast cancer. It serves as a transcriptional coregulator, interacting with estrogen receptor (ER), androgen receptor (AR), and, in endometrial cancer, the progesterone receptor (PR). SRARP inhibits cell proliferation, migration, and invasion via suppression of the epithelial-mesenchymal transition (EMT), largely through negative regulation of the non-canonical Wnt signaling pathway. Loss or silencing of SRARP correlates with worse outcome in several malignancies, and its overexpression demonstrates tumor suppressor activity. SRARP’s biological activity depends on its role in the transcriptional regulation of steroid receptor signaling, making it a candidate biomarker and potential therapeutic target in hormone-sensitive cancers.
