Overview

Sterol regulatory element-binding protein 1 (SREBP-1) is a key transcription factor that regulates the expression of genes involved in lipid biosynthesis, including fatty acid, triglyceride, and cholesterol synthesis. It plays a central role in maintaining lipid homeostasis and is implicated in various metabolic diseases, including obesity, type II diabetes mellitus, and non-alcoholic fatty liver disease. Upregulated activity also supports tumor cell growth. There are two major isoforms, SREBP-1a and SREBP-1c, which differ in their tissue distribution and specific functions in lipid metabolism.

Mechanism of action

SREBP-1 is a transcription factor that binds to sterol regulatory elements (SREs) within promoters of target genes to regulate their expression. Drugs can target the activation/cleavage process, nuclear translocation, or DNA binding activity of SREBP-1 to modulate lipid metabolism.
