Overview

Sterol Regulatory Element-Binding Protein 2 (SREBP2) is a transcription factor that primarily regulates genes involved in cholesterol biosynthesis and uptake. It is anchored to the endoplasmic reticulum when intracellular cholesterol levels are high and is released and activated when cholesterol levels decrease. Upon activation, SREBP2 is cleaved by site-1 protease (S1P) and site-2 protease (S2P), releasing the NH2-terminal fragment (nSREBP2), which then translocates to the nucleus to activate target genes. Variants of the SREBF2 gene have been associated with an increased risk of knee osteoarthritis, and SREBP2 is being explored as a potential target for therapies related to lipid metabolism disorders, such as non-alcoholic steatohepatitis (NASH).
