Overview

Sterol regulatory element-binding transcription factor 1a (SREBP-1a) is an isoform of SREBP-1, a key transcription factor regulating genes involved in lipid and cholesterol synthesis. SREBP-1a originates from the SREBF1 gene by alternative promoter usage and is broadly expressed, including in the intestine and spleen. It exerts its function by binding sterol regulatory elements (SREs) in the promoters of genes controlling lipid biosynthesis, thus playing a central role in metabolic regulation. SREBP-1a is synthesized as an ER/nuclear membrane-bound precursor and is activated via proteolytic cleavage in response to low sterol levels, releasing the active domain into the nucleus. Dysregulation of SREBP-1a is implicated in diseases characterized by altered lipid metabolism, including non-alcoholic fatty liver disease, metabolic syndrome, and various cancers[1][2][5].
