Overview

Stimulator of chondrogenesis 1 (SCRG1) is a small, cysteine-rich protein encoded by the *SCRG1* gene, originally identified due to its upregulation in neurodegenerative conditions linked to prion infection[1][3][4]. It is predominantly expressed in the central nervous system, articular cartilage, aorta, and testes, and is secreted or membrane-associated due to an N-terminal hydrophobic signal sequence[1][2][3]. In cartilage biology, SCRG1 promotes the differentiation of mesenchymal stem cells toward a chondrogenic phenotype and is upregulated during cartilage formation and regeneration[2][5]. Mechanistically, SCRG1 interacts with bone marrow stromal cell antigen 1 (BST1), and enhances chondrogenesis by inhibiting the canonical Wnt/β-catenin signaling pathway through upregulation of Wnt5a[2]. In osteoarthritis, SCRG1 expression is elevated in inflamed synovium and articular cartilage, and it appears to be a promising biomarker for disease activity and a potential target for therapeutic intervention[4]. Additionally, SCRG1 regulates immune-related genes and pathways in osteoarthritic tissue, participates in host response to neurodegeneration, and associates with secretory vesicles in neurons[3][4]. There are no approved drugs that directly target SCRG1, but bioinformatic analyses suggest possible modulation by compounds such as valproic acid[4].
