Overview

The Stimulator of interferon genes 1 (STING1) gene promoter, also known as the TMEM173 promoter, is a critical regulatory DNA sequence that controls the expression of the STING protein, a central mediator of the innate immune response to cytosolic DNA (Source: Wikipedia, 2024). In various malignancies, including melanoma and glioma, the STING promoter frequently undergoes epigenetic silencing through hypermethylation of CpG islands, which prevents the production of STING and allows tumors to evade immune surveillance (Source: NIH, 2022). This silencing can be therapeutically reversed using DNA methyltransferase inhibitors (DNMTis) such as decitabine and azacitidine, which promote promoter demethylation and restore STING-mediated signaling (Source: ResearchGate, 2022).
