The STING (Stimulator of Interferon Genes) signaling pathway is a central component of the innate immune response, primarily responsible for detecting cytosolic DNA and initiating downstream immune signaling. The pathway involves cGAS activation by cytosolic DNA, leading to cGAMP production. cGAMP binds to STING, activating it and triggering translocation from the ER to the Golgi. This leads to TBK1 activation, IRF3 phosphorylation and nuclear translocation, and NF-kB activation, resulting in type I IFN and inflammatory cytokine production. Dysregulation is linked to various diseases.
