Stomatin-like protein 1 (STOML1) is a member of the stomatin protein family, predominantly expressed in brain, heart, and skeletal muscle, but detectable in other tissues as well[1][5]. It is a membrane-associated protein with a bipartite structure: an N-terminal stomatin domain and a C-terminal sterol carrier protein-2 (SCP-2) domain[1][2]. STOML1 primarily localizes to late endosomes, where it participates in cholesterol/sterol transfer and associates with detergent-resistant membranes (lipid rafts). It interacts directly with stomatin and plays a role in the formation of cholesterol-rich endosomal vesicles when cholesterol trafficking is impaired[1][2]. STOML1 can modulate acid-sensing ion channels (notably inhibits ASIC1 currents), and may play a regulatory role in neuronal signaling[3]. While implicated in lipid homeostasis and ion channel regulation, there are no known drugs directly targeting STOML1, and it is not currently considered a standard therapeutic target in clinical use.
