Stonin-1 (STON1) is an endocytic adaptor-like protein and one of two human homologs of the Drosophila stoned B protein[1][4][5]. It is involved in the clathrin-mediated endocytosis of selected cell surface proteins, with a modular structure comprising an N-terminal proline-rich domain, a central region of stoned B-homology, and a C-terminal region homologous to the mu subunits of adaptor protein (AP) complexes[1][4]. STON1, as part of the endocytic machinery, supports vesicle formation at the plasma membrane and is essential for presynaptic vesicle cycling in neurons (based on homology and studies in Drosophila). In humans, its roles expand to vesicle trafficking and cellular signaling. Recent research points to potential involvement in the modulation of the tumor immune microenvironment, particularly in kidney renal clear cell carcinoma, and genetic studies suggest a connection to polycystic ovary syndrome, though precise mechanisms and disease associations require further study[1]. STON1 is not currently classified as a classical drug target (such as a receptor, enzyme, or transporter), and there are no known therapeutic drugs directly targeting this protein[1][4].
