Overview

The **store-operated calcium entry channel protein** commonly refers to Orai1, an integral plasma membrane protein that forms the pore of Calcium release-activated calcium (CRAC) channels, mediating store-operated calcium entry (SOCE) into cells. Orai1 is activated when depletion of endoplasmic reticulum (ER) Ca²⁺ stores is sensed by ER-resident proteins named STIM1 and STIM2. Upon store depletion, STIM proteins undergo conformational change and translocate to ER–plasma membrane junctions, where they physically interact with Orai1, triggering channel opening and allowing extracellular calcium influx. This mechanism is essential for diverse biological processes including signal transduction, gene expression, immune cell activation, and maintenance of cellular calcium homeostasis. Orai1 dysfunction is implicated in immunodeficiency, muscle disorders, and neurological diseases. Targeting the Orai1 channel and SOCE pathway is under investigation for immunomodulation and treatment of pathological calcium signaling in various diseases[1][2][3][5][6].
