Overview

Store-operated calcium entry regulator STIMATE (STIMATE, also known as TMEM110) is an endoplasmic reticulum-resident multi-transmembrane protein that regulates store-operated calcium entry (SOCE) by maintaining and remodeling ER-plasma membrane junctions crucial for calcium signaling. It acts as a positive modulator of SOCE by physically associating with stromal interaction molecule 1 (STIM1), promoting STIM1’s conformational switch to an activated state and facilitating the interaction with ORAI1 calcium channels on the plasma membrane. Depletion of STIMATE/TMEM110 impairs STIM1 relocalization, reduces Ca²⁺ influx, and suppresses downstream activation of the calcineurin/NFAT signaling pathway. This protein is not a classic therapeutic target (e.g., receptor, enzyme, or transporter) but is important for the physiological regulation of calcium signaling complexes at the ER–plasma membrane interface
