Overview

Storkhead-box protein 2 (STOX2) is a member of the winged-helix domain-containing transcription factors and is closely related to STOX1[1][2]. It is differentially expressed in decidual tissue and plays a crucial regulatory role in placental development, specifically impacting susceptibility to pre-eclampsia with fetal growth restriction[1]. STOX2 is an integral component of the TGF-β signaling network in human embryonic stem cells, where it modulates the stability and abundance of SMAD2 and SMAD4 proteins, thereby influencing the expression of classical TGF-β target genes[2]. Dysregulation or mutation of STOX2—including its cis-regulatory long non-coding RNA—can disrupt normal trophoblast differentiation and contribute to early-onset pre-eclampsia and related syndromes[1]. Emerging research also links STOX2 to cancer biology, such as promoting oral squamous cell carcinoma progression[1]. Currently, STOX2 is not a direct drug target, but its involvement in developmental pathways and disease suggests potential significance in biomarker discovery and mechanistic understanding of relevant pathologies[1][2].
