The stratum corneum barrier function refers to the protective role of the outermost layer of the skin. This function is achieved through a complex structure of corneocytes (protein-rich cells) held together by a lipid matrix composed of ceramides, cholesterol, and free fatty acids. The barrier prevents excessive water loss, blocks entry of pathogens and toxins, maintains hydration via natural moisturizing factors (NMF), and contributes to immune defense through physical separation and antimicrobial peptides. Impairment of this function is associated with skin diseases like atopic dermatitis, psoriasis and ichthyosis.
