This complex matrix is predominantly composed of ceramides, cholesterol, and free fatty acids organized into highly ordered lamellar structures, specifically the long periodicity phase and short periodicity phase (PubMed: PMCID PMC2835894). Its fundamental biological function is to prevent excessive transepidermal water loss (TEWL) and protect the body from environmental insults, including pathogens and chemical toxins (PubMed: PMCID PMC3105619). In various dermatological conditions such as atopic dermatitis and psoriasis, the composition and organization of these lipids are significantly altered, leading to barrier dysfunction and chronic inflammation (PubMed: PMCID PMC4252662).
