Target intelligence / Profile preview
The stratum corneum intercellular lipids are a specialized lipid matrix located in the extracellular spaces of the skin's outermost layer, the stratum corneum. This matrix is primarily composed of ceramides, cholesterol, and free fatty acids, which are organized into dense, crystalline lamellar structures that provide the skin's essential permeability barrier (Source: NIH PMC2843412). Their primary biological function is to regulate transepidermal water loss (TEWL) and prevent the entry of environmental pathogens and allergens (Source: StatPearls NBK441980). In pathological states like atopic dermatitis and psoriasis, the ratio and structural organization of these lipids are often disrupted, leading to impaired barrier function and increased susceptibility to inflammation (Source: PubMed 24656726). Therapeutic strategies target these lipids through the application of barrier-repair emollients containing physiological lipid mixtures to restore the skin's integrity. Additionally, these lipids are a major consideration in transdermal drug delivery, as they represent the primary rate-limiting barrier for the absorption of topical medications.
Restoration of the lipid lamellar structure through replenishment of physiological lipids and physical occlusion to reduce transepidermal water loss.
