Protease activity in the stratum corneum (SC) refers to the enzymatic breakdown of proteins within this outermost layer of the skin. This process is essential for normal skin desquamation (shedding of corneocytes), maintenance of barrier function, and overall skin homeostasis. Key enzymes include kallikrein-related peptidases (KLKs), tryptase, and SASPase/cathepsin D. Regulation is primarily mediated by endogenous inhibitors like LEKTI and pH-dependent activation.
