Overview

Skin barrier proteins are a group of structural and functional proteins primarily located in the stratum corneum (SC) that maintain the integrity, hydration, and protective functions of the skin. They prevent transepidermal water loss (TEWL) and protect against external insults such as pathogens, allergens, and chemicals. Key proteins include filaggrin (FLG), involucrin, loricrin, small proline-rich proteins (SPRPs), keratins, and desmosomal proteins like desmoglein 1 (DSG1) and desmocollin 1 (DSC1). Mutations or dysregulation of these proteins can lead to compromised barrier function and skin diseases like atopic dermatitis and ichthyosis vulgaris.
