The stratum corneum is the skin’s outermost layer, composed mainly of corneocytes surrounded by lipids, with the cornified cell envelope formed from aggregated and cross-linked structural proteins such as keratin and filaggrin. These proteins provide strength, flexibility, and water impermeability, enabling the skin to act as a robust barrier against physical, chemical, and microbial threats. Filaggrin also generates natural moisturizing factors, while proteins like involucrin and loricrin reinforce the corneocyte envelope and regulate desquamation. Defects in these structural proteins (notably filaggrin mutations) are strongly linked to skin diseases, especially atopic dermatitis and other barrier-impairment disorders. No single molecule in this class serves as a canonical drug target, but these proteins jointly determine the stratum corneum’s resilience, hydration, and defense capacities.
