Strawberry notch homolog 1 (SBNO1) is a highly conserved protein classified as a chromatin regulator in the SF2 helicase superfamily[2][4]. It contains characteristic DExD/H-box and helicase C-terminal domains, conferring putative DNA and histone binding abilities[4][7]. SBNO1 acts exclusively in the nucleus, where it regulates transcription, especially enhancer activation for lineage-defining genes like Cdx2 during early mouse development[2][3]. It integrates mechanical and signaling cues from the Notch and Hippo pathways, enabling robust transcriptional activation in trophoblast cells; SBNO1 knockout results in failed blastocyst formation and embryonic lethality[2][3]. SBNO1 also modulates neural stem cell proliferation and neuroinflammatory responses via PI3K-Akt and Wnt/β-catenin signaling[5][1]. Although implicated in prostate cancer and muscular dystrophy, direct clinical targeting or biomarker use is not established. Interactions include physical association with chromatin modifiers such as FACT, and signaling transcription complexes like Yap/Tead4, and Notch/Rbpj[2]. SBNO1’s precise mechanisms remain a subject of ongoing research, and its function underlines the intersection of chromatin remodeling, developmental signaling, and lineage specification[2][3][5].
