Overview

Streptococcus pneumoniae capsular polysaccharide–protein conjugate refers to the chemical complex formed by covalent linkage of purified capsular polysaccharides from selected serotypes of S. pneumoniae to an immunogenic carrier protein, such as CRM197 or diphtheria toxoid. This conjugation transforms the polysaccharide from a T cell–independent antigen into a T cell–dependent immunogen, enabling young children and other poorly responsive populations to generate strong, durable antibody responses and immunological memory. The conjugate itself is not a natural biomolecule or drug target in the classical pharmacological sense, but rather a designed antigenic complex used exclusively in the context of pneumococcal conjugate vaccines for prevention of invasive pneumococcal disease and pneumonia by inducing protective, serotype-specific immunity through antibody production and opsonophagocytosis[1][2][3][4][5][6][7][8].

Mechanism of action

When administered as a vaccine, the conjugated polysaccharide is recognized by B cells, internalized, and presented with carrier protein peptides to T cells, eliciting a strong T cell-dependent antibody response. It induces formation of memory B cells and durable immune protection, and promotes opsonophagocytic killing of Streptococcus pneumoniae.
